背景:一氧化氮(NO)在大量生物学功能中起着重要的调节作用,因此具有极大的潜力,可用于临床应用。然而,纳米 - 一氧化氮递送系统的分子设计中的化学物质目前还缺乏。 目的:本综述的总体目标是为读者提供与NO释放分子(NORMs)的设计,加载和释放机制以及为纳米治疗药物递送NORM有关的基础知识。 方法:我们对NORMs的设计和合成以及当前最先进的NO兼容递送平台进行了全面的文献检索,以满足各种临床需求。 结果:用于抗癌和抗微生物应用的N-diazeniumdiolate和基于S-亚硝基硫醇的NO分子是应用最广泛的NORM。这些NORM与细胞相容性有机和无机纳米载体的创新整合使得能够控制目标病变部位的时空递送和NO的释放。结论:我们全面总结了支持NORMs分子设计的基础化学物质,并批判性评估了纳米NO输送系统在先进生物医学应用方面的最新进展。

摘要: Background: Nitric oxide (NO) plays important regulatory roles in a plethora of biological functions and thus holds tremendous potential to be exploited for clinical uses. However, the chemistries in the molecular design of nano-nitric oxide delivery systems is currently lacking.
Objective: The overarching aim of this review is to provide the readers with the fundamentals that relate to the design of NO release molecules (NORMs), loading and releasing mechanism, as well as delivery of NORMs for nanotherapeutics.
Methods: We conducted a thorough literature search on the design and synthesis of NORMs, as well as the current state-of-the-art NO compatible delivery platforms to address various clinical needs.
Results: N-diazeniumdiolate and S-nitrosothiol based NO molecules are among the most widely used NORMs for anti-cancer and anti-microbial applications. The innovative integration of these NORMs with cytocompatible organic and inorganic nanocarriers enabled controlled spatiotemporal delivery and release of NO at the targeted diseased sites.
Conclusion: We have provided a comprehensive summary of the fundamental chemistries underpinning the molecular design of the NORMs and critically assessed the recent advancements of nano-NO delivery systems for advanced biomedical applications.
