左旋肉碱及其酯的质谱分析:肉碱稳态中潜在的生物标志物

目的:在三十年前经典肉碱研究的黄金时代之后,质谱的普及开辟了新的视角,如今对肉碱系统有了更好的了解。在经典时期,一些人和动物研究集中于该分子的各种不同生理功能,这些研究揭示了肉毒碱在正常和病理条件下体内稳态的不同方面。最初,实验室分析是基于经典或放射性酶分析方法,仅能测定游离和总肉碱水平,并能计算出总肉碱酯含量,而无需任何酰基组成信息。质谱法的引入使得可以测定游离肉碱,并可以灵敏地测定不同的肉碱酯。除了基础研究之外,肉碱酯的质谱研究还被引入新生儿筛查程序中,因为它能够同时检测30多种代谢异常。此外,进行了质谱测量以研究影响肉碱稳态的不同疾病状态,例如糖尿病,慢性肾衰竭,腹腔疾病,心血管疾病,自闭症谱系障碍或炎症性肠病。 结果:本文将回顾肉碱研究领域在正常和各种病理状态下酰基肉碱的质谱分析方面的最新进展。 结论:越来越多的出版物使用质谱分析作为研究正常生理状况或揭示原发性和继发性肉碱缺乏症的潜在生物标志物的工具,表明该工具为肉碱研究提供了新视角。
