免疫系统积极地抵消肿瘤的发生过程;免疫系统的功能或识别转化细胞的能力的突破,可能会促进癌症的发展。癌症通过多种机制得以逃脱免疫系统的控制,这些机制目前仅在细胞和分子水平上被部分了解。在这些机制中,在过去的十年中,所谓的“抑制性免疫检查点”的作用正在成为阻止免疫系统攻击肿瘤的关键。在生理上,抑制性免疫检查点的作用是维持机体的自耐受性和减轻致病性感染引起的组织损伤。肿瘤细胞利用这些免疫抑制分子来对比免疫干预和诱导肿瘤耐受。针对这些检查点的分子制剂代表了癌症治疗的新前沿。尽管肿瘤之间分子改变的异质性和多样性,免疫检查点靶向治疗已被证明对选择的甚至是组织学上不同类型的癌症有帮助,并且目前正在被用于对抗越来越多的肿瘤。最常用的是基于MOAB的免疫疗法,它针对程序性细胞死亡蛋白1(PD-1)、PD-1配体(PD-L1)或细胞毒性T淋巴细胞抗原-4 (CTLA4)。然而,随着癌细胞利用新的免疫检查点的发现,新的治疗方法目前正在开发中。本文就目前已知的抑制检查点以及肿瘤免疫编辑的机制进行综述。概述免疫检查点靶向方法,也包括联合免疫疗法和现有的试验,也提供。尽管研究人员在反应的生物标记物领域付出了巨大的努力,但到目前为止,还没有验证过FDA批准的用于癌症患者的免疫生物标记物。我们强调了预测生物标志物的相关研究,并试图讨论这一领域的挑战,因为驱动肿瘤免疫耐受的动态机制复杂且大部分未知。
