紫杉醇(PTX)是一类一线药物,用于无法从靶向治疗或免疫治疗中受益的晚期非小细胞肺癌(NSCLC)患者。然而,长期治疗患者总是会产生对PTX的耐药性。尽管在文献中已充分证明了导致PTX抗药性的各种机制,但基于这些机制克服NSCLC中PTX抗药性的策略仍具有挑战性。在本文中,我们综述了阐明NSCLC中PTX耐药性的主要机制的最新进展,包括ABC转运蛋白的过表达,微管蛋白结构的改变以及细胞因子,miRNA,激酶信号通路和上皮-间充质转化的参与。还讨论了可能有助于指导治疗决策并恢复细胞对PTX敏感性的PTX耐药性或PTX反应的潜在标志物。最后,我们总结了克服NSCLC细胞中PTX耐药性的相应策略,这可能为临床试验提供新见解,并在将来使肺癌患者受益。
