摘要
几十年来,中性粒细胞通常被认为是具有促炎和吞噬特性的先天免疫细胞,具有双重活性,有益(抗菌)和有害(组织损伤)。重要的是,在Toll样受体(TLRs)发现之前,中性粒细胞在适应性免疫中的作用仅限于体液反应和调控抗原的吞噬效应阶段。此外,一般认为中性粒细胞和整个先天免疫系统在功能上与适应性免疫无关。在我们证明用HOCl氯化蛋白时,中性粒细胞MPO-卤化系统的主要产物增强了蛋白的免疫原性。基于这一发现,我们首次提出了中性粒细胞作为APC辅助细胞参与适应性免疫诱导阶段的新作用。此后,我们发展了中性粒细胞作为连接先天免疫和适应性免疫的细胞的作用的理论。我们提出,HOCl的蛋白修饰可能作为一个嗜中性粒细胞依赖的分子标记系统,通过它哨点树突状细胞可以更快地识别病原体来源的抗原。同时,研究表明,牛磺酸是中性粒细胞胞浆中最丰富的游离氨基酸,也是HOCl的主要清除剂,是氧化-抗氧化网络的一部分,负责调节和终止急性炎症。此外,牛磺酸氯胺(TauCl)是HOCl与牛磺酸反应的生理产物,具有抗菌和抗炎作用。本文将讨论HOCl、牛磺酸和牛磺酸在先天性免疫和适应性免疫中的作用。
