摘要
先前的研究已经表明,阿尔茨海默病(AD)和遗忘型轻度认知障碍(aMCI)显示前扣带皮层(ACC)解剖和功能异常,越来越多的证据支持ACC的变化可以预测从aMCI到AD的进程这一假说。在本研究中,我们旨在探讨aMCI如何改变这两个功能和结构异构的ACC的子区域即背侧ACC(dACC)和腹侧ACC(vACC)的功能连接;结构连接是否会影响两个ACC子区域之间的功能连接。为探索这一假说,我们研究了静息状态功能下24例 aMCIs组和29例健康对照组的fMRI和DTI图像。dACC的功能连接在显性网络(SN)明显增强;vACC的功能连接减弱。DTI结果表明,aMCI双侧扣带回纤维受损最严重的区域中,左前扣带部分畸形与两侧ACC子区域的功能连接显著相关。总之,本研究显示ACC在SN内在功能连接的病理学改变以及aMCI中dACC和vACC间的联系。我们的研究也显示,破坏白质完整的扣带回前部区域与ACC子区域连通性的改变有关。

Title:Disrupted Functional Connectivity Related to Differential Degeneration of the Cingulum Bundle in Mild Cognitive Impairment Patients

摘要: Previous studies have demonstrated that Alzheimer’s disease (AD) and amnestic mild cognitive impairment (aMCI) exhibited anatomical and functional abnormalities in the anterior cingulate cortex (ACC) and accumulating evidence supported the hypothesis that changes in the ACC predict the progression from aMCI to AD. In this study, we aimed to explore how the two functional and structural heterogeneous sub-regions of ACC, namely the dorsal ACC (dACC) and the ventral ACC (vACC), changed in aMCI and whether the structural connectivity affects the functional connectivity between the two ACC subregions. To investigate this hypothesis, we studied resting-state fMRI and DTI images in a group of 24 aMCIs and 29 healthy controls. The dACC exhibited a significantly increased functional connectivity in the Salience Network (SN) and a decreased functional connectivity with the vACC in aMCI. The DTI results showed that the bilateral cingulum fibers were the most damaged tracts in aMCI and that the fractional anisotrophy of the left anterior cingulum was significantly correlated with the functional connectivity between the two ACC sub-regions. In conclusion, this study revealed the pathological changes in the intrinsic functional connectivity of the ACC within SN, as well as the connectivity between the dACC and vACC in aMCI. Our study also revealed that disrupted white matter integrity of the anterior regions of the cingulum was associated with the alterations in subregional connectivity in the ACC.
