摘要
尽管经过了几十年的研究和努力,仍然没有有效的治疗阿尔茨海默病(AD)的疾病改善疗法。最近的AD临床试验大多以淀粉样蛋白途径为靶点,但均以失败告终。虽然淀粉样病变是AD的标志和定义特征,但由于疗效低、副作用严重,靶向淀粉样途径一直是一个很大的挑战。我们了解AD中记忆丧失的主要原因的替代方法或机制需要被视为潜在的治疗靶点。越来越多的研究表明,Ca2+失调在AD病理中发挥重要作用,并与其他AD异常相关,如过度炎症、ROS增加、自噬受损、神经退行性变、突触和认知功能障碍。在各种AD模型中,细胞质空间、内质网(ER)和线粒体中的Ca2+失调已被报道。药物或策略,纠正Ca2+失调的AD,已被证明是一种有前途的方法治疗AD的临床前模型。这篇综述将讨论Ca2+失调在AD中的机制和相关病理,并讨论通过靶向Ca2+失调开发治疗AD的新药的潜在方法或策略。
