摘要
前列腺癌是影响世界各地男性人口的第二大常见癌症。脂质代谢紊乱与前列腺癌的相关性早已为人们所熟知。根据假说,胆固醇可能通过Akt信号转导和新甾体生成等多种生物学机制参与前列腺生长和分化的信号分子,从而导致前列腺癌进展。一些研究的结果表明,胆固醇水平的增加可能与更严重的疾病病程的高风险有关。上述脂肪酸合成的改变是癌症的一个独特特征,因此,成为前列腺癌治疗干预的一个有吸引力的目标。药物或基因治疗旨在降低参与脂肪酸从头合成的酶的活性,特别是FASN、ACLY (ATP柠檬酸裂解酶)或SCD-1(硬脂酰辅酶A去饱和酶),这可能导致细胞生长停滞。然而,并不是所有的癌症都与低胆固醇血症明确相关。不能排除前列腺癌和脂质紊乱之间的关系并不是癌变和循环胆固醇量之间的直接定量关系。也许对应关系更为复杂,并与胆固醇组分甚至高密度脂蛋白胆固醇的子组分的分布有关。
