FAM13A、PHLDB1和CYP24A1多态性在乳腺癌风险中的作用

背景:单核苷酸多态性(SNPs)是乳腺癌易感性的重要指标. 目的:探讨中国汉族人群FAM13A、PHLDB 1和CYP24A1基因SNP与乳腺癌发病的关系。 方法:对379例女性乳腺癌患者和407例女性健康对照者进行病例对照研究。用Agena MassARRAY平台对3个SNP进行了基因分型使用χ2检验比较病例组和对照组之间的多态性等位基因和基因型频率。 遗传模型分析通过使用逻辑回归计算比值比(OR)和95%置信区间(CI)来评估SNP与乳腺癌风险之间的关联。 RegulomeDB和HaploReg数据库用于计算多态性的可能的功能效果。 结果:总体分析结果表明,rs4809957与乳腺癌风险增加相关(等位基因A:OR = 1.27,95%CI:1.03-1.55,p = 0.024; AA vs. GG:OR = 1.80,95%CI: 1.15–2.82,p = 0.010;隐性模型:OR = 1.70,95%CI:1.12-2.58,p = 0.012); 在隐性模型中发现rs1059122与降低乳腺癌风险有关(OR = 0.71,95%CI:0.51-0.98,p = 0.039)。 分层分析发现三个SNP(rs1059122,rs17748和rs4809957)与乳腺癌风险之间存在显着关联。 结论:我们的结果表明,rs1059122(FAM13A),rs17748(PHLDB1)和rs4809957(CYP24A1)可能对中国汉族人群的乳腺癌易感性有贡献。 需要进一步的大样本研究来证实我们的发现,还需要进行功能研究以探索其在乳腺癌发展中的作用。
