摘要
背景:一氧化氮(NO)是关键信号分子,在炎症中具有重要作用。它可以由内皮细胞,嗜中性粒细胞和其他细胞分泌,一旦循环,NO在调节各种嗜中性粒细胞的细胞活动和命运中起着重要作用。 目的:描述NO及其伴随的过氧亚硝酸盐对中性粒细胞的细胞反应及中性粒细胞凋亡的信号传导机制。 方法:回顾文献以评估NO对中性粒细胞的影响。 结果:NO在各种嗜中性粒细胞的细胞活动以及与其他细胞的相互作用中起着重要作用。中性粒细胞的特征性细胞活性是粘附和吞噬作用。 NO通过阻止嗜中性粒细胞粘附和活化的嗜中性粒细胞破坏内皮细胞而在嗜中性粒细胞-内皮相互作用中起保护作用。 NO抑制中性粒细胞的吞噬活性,但通过肾小管小体延伸或细胞因子刺激长距离接触相互作用。中性粒细胞是超氧化物的主要来源,但NO流动导致过亚硝酸盐的形成,过氧亚硝酸盐是一种具有高生物活性的化合物。过氧亚硝酸盐参与类二十烷酸生物合成的调节并抑制内皮前列环素合酶。 NO和过亚硝酸盐调节细胞5-脂氧合酶活性和白三烯合成。嗜中性粒细胞长期暴露于NO会导致细胞死亡机制和嗜中性粒细胞凋亡的激活。 结论:一氧化氮和一氧化氮/一氧化氮相互作用调节中性粒细胞生命和死亡的微调机制。
