自上个世纪以来,当科学家提出锁和钥匙模型时,药物的发现就集中在作用于单一目标的药物的开发上。但是,单目标药物疗法对具有多因素发病机制的复杂疾病无效。而且,单一靶标药物的组合容易引起耐药性和副作用。近年来,在FDA批准的药物中越来越多地使用多目标药物。阿尔茨海默氏病(AD)是一种复杂的多因素疾病,其确切的分子机制仍未完全了解。近年来,结合多种药物的药效基团的合理的多靶点药物设计方法已越来越多地用于抗AD药物的开发中。在这篇综述中,我们简要介绍了AD的发病机理,并提供了有关具有多个靶标(例如淀粉样β肽,Tau蛋白)的抗AD药物化学结构(2013年至今)的最新进展的详细讨论。 ,胆碱酯酶,单胺氧化酶,β位淀粉样前体蛋白裂解酶1,自由基,金属离子(Fe2 +,Cu2 +,Zn2 +)等。在本文中,我们还添加了一些新的靶标或可能的发病机理,这些新靶标或可能的发病机理近年来已报道用于AD治疗。我们希望这些发现可以为AD的药物治疗提供新的观点。
