摘要
背景:积累的实验数据支持天然化合物干预某些人类发病机制中复杂的分子途径的能力。其中,糖尿病是当今世界上与死亡风险增加有关的流行病。因此,检测新型抗糖尿病药和/或佐剂至关重要。生物碱代表具有多种治疗特性的多种天然产物。在过去的20年中,有关其抗糖尿病能力的已发表的研究大大增加了。 目的:讨论当前有关某些生物碱抗糖尿病作用的概念,并特别提及它们在整个胰岛素信号通路中的分子靶标。 方法:在SCOPUS和PUBMED数据库中进行深入搜索后,对具有胰岛素分泌/致敏特性的生物碱的文献进行了严格审查。 结果:体内和体外的证据支持小ber碱,trigonelline,胡椒碱,氧化苦参碱,芬多林,evodiamine和neferine对β细胞,心肌细胞,脂肪细胞,肝细胞和其他细胞中胰岛素信号传导和相关级联的影响。相关的受体,激酶,激素和细胞因子在基因转录,蛋白质表达,活性和/或磷酸化方面受到影响。与胰岛素抵抗,β细胞衰竭,氧化应激和炎症以及临床表型有关的病理生理过程也受到影响。 讨论:越来越多的证据表明特定生物碱具有干预胰岛素信号转导途径的能力,逆转导致胰岛素抵抗和葡萄糖耐受不良的分子缺陷并改善体内和体外疾病并发症的能力。预计未来的深入分子研究将阐明其确切的作用机理,同时迫切需要进行大规模的临床试验来评估其作为抗糖尿病药的潜力。
