摘要
阿尔茨海默病(AD)是一种渐进的多因素神经退行性疾病。目前,没有有效的治疗方法,这是由于多种因素涉及的病理生理学和严重程度的AD。最近,一种新药物候选药物的合理设计方法,也被称为多目标导向配体(MTDL)策略,已被用于开发多种能够同时作用于多种生物靶点的混合化合物。发现能够针对AD发病机制的多种因素的药物候选物将极大地促进治疗策略的改进。本综述是对我们小组最近发表的另一篇综述文章的补充,该文章涵盖了2005-2012年的前一时期,并着重介绍了MTDLs方法在合理设计治疗AD的新药候选原型方面的最新进展和实例。

摘要: Alzheimer’s disease (AD) is a progressive multifactorial neurodegenerative disorder. Currently, no effective treatment is available and this is due to multiple factors involved in pathophysiology and severity of AD. A recent approach for the rational design of new drug candidates, also called multitarget-directed ligands (MTDL) strategy, has been used to develop a variety of hybrid compounds capable to act simultaneously in diverse biological targets. The discovery of drug candidates capable of targeting multiple factors involved in AD pathogenesis would greatly facilitate in improving therapeutic strategies. This review is a complement to another review article, recently published by our group, which covered the previous period of 2005-2012, and highlights recent advances and examples of the exploitation of MTDLs approach in the rational design of novel drug candidate prototypes for the treatment of AD.
