Abstract
Background: The weak water solubility is the main problem to develop the highly potent pharmaceuticals. Nowadays to improve the solubility of various formulations containing nanocrystals, specific strategies are involved for their dissolution and bioavailability.
Objective: The objective of the present work was to improve the solubility of low watersoluble aceclofenac by preparing stable nanocrystals. Methods: The methods for preparing the aceclofenac nanocrystals was carried out with the help of combination of precipitation process by using antisolvent and high pressure homogenization required for stability and converting them into fine and dry crystals by using spray-drying. The drug and its nanocrystals were characterized for SEM, FT- IR and XRD as well as measuring the particle size. Results: The XRD results indicated that the antisolvent precipitation with high-pressure homogenization process led to the crystallization of aceclofenac. The nanocrystals thus produced lead to the stabilization of drugs by adsorption of the stabilizers onto the drug surface. Aceclofenac nanocrystals increased the solubility of drug by almost fourfold. The combination of two methods for forming nanocrystals leads to the formation of a stable formulation which showed increase in the solubility by increment in effective surface area and amorphous nature of drug.Keywords: Nanocrystals, aceclofenac, solubility, particle size, drug, homogenization, amorphous.
