NF-κB1内含区多态性是巴基斯坦人乳头瘤病毒感染人群头颈癌的危险因素

摘要

背景:头颈癌(HNC)的发生有多种危险因素,包括人乳头瘤病毒(HPV)感染。遗传易感性也在解除对不同信号通路的调控中发挥重要作用,包括NF-KB通路。某些多态性被报道影响NF-kB通路基因。目的:本研究旨在研究巴基斯坦HNC患者中HPV与NF-KB1 (p50)基因多态性的关系。方法:用Exgene SV DNA提取试剂盒提取HNC肿瘤的基因组DNA。对NF-κB1 SNPs 94ins/del (rs28362491)、rs1598858和rs4648068进行等位基因特异性PCR和直接测序分析。结果:AGrs1598858、AGrs4648068和GGrs4648068基因型与研究人群头颈癌风险显著增加相关。此外,AGrs1598858和GGrs4648068基因型的HNC病例表现出HPV相关癌症的风险增加。在研究人群中未检测到启动子区域SNP 94ins/del (rs28362491)。吸烟、淋巴结浸润和低分化肿瘤与HPV诱导的癌症呈正相关。结论:这是巴基斯坦首次对NF-κB1基因多态性进行全面的研究。NF-κB1基因AGrs4648068、GGrs4648068和AGrs1598858基因型与巴基斯坦研究的HPV感染人群头颈部癌症风险增加相关。研究结果表明,HPV感染、淋巴结浸润和烟草使用对HPV诱导的HNC具有协同作用,并通过内含子SNPs的NF-κB1基因调控HNC。
