摘要
乳腺癌是全世界女性最常见的恶性肿瘤。传统的治疗方法,包括放射治疗和内分泌治疗,乳腺癌治疗后都有不可避免的副作用。近几十年来,靶向治疗乳腺癌的方法发展迅速,前景广阔。JAK/STAT信号通路在乳腺癌的发生、发展和转移中发挥重要作用。因此,人们开发了许多JAK和STAT蛋白的小分子抑制剂。这些抑制剂在细胞和动物模型中都显示出对乳腺癌的有效抑制作用,甚至其中一些已经在临床试验中。本文就JAK/STAT信号转导途径在乳腺癌发病机制中的作用以及JAK/STAT抑制剂在乳腺癌治疗中的应用前景作一综述。

摘要: Breast cancer is the most common malignant tumor in women worldwide. Traditional ways of treatment, including radiotherapy and endocrine therapy, for breast cancer have inevitable side effects. In recent decades, targeted therapies for breast cancer have rapidly advanced and shown a promising future. The JAK/STAT signaling pathway has been shown to play important roles in tumorigenesis, maintenance and metastasis of breast cancer. Hence, many small molecule inhibitors of JAK and STAT proteins have been developed. These inhibitors exhibit potent inhibitory effects on breast cancer in both cellular and animal models, and even some of them have already been in clinical trials. This review article discussed the JAK/STAT signal transduction pathway in the pathogenesis of breast cancer, and the potential for the application of JAK/STAT inhibitors in breast cancer treatment.
