背景:心血管疾病(CVD)仍然是全球范围内主要的死亡原因。如今,CVD治疗中的精准医学概念要求结合个体的遗传和环境变异性,以实现个性化的疾病预防和量身定制的治疗。自噬是进化上保守的细胞内降解过程,已被证明在各种CVD的发病机理中必不可少。但是,目前还没有针对自噬性CVD的有效治疗方法。长非编码RNA(lncRNA)是非编码RNA序列,在自噬调节中发挥多种作用,但是,关于lncRNA与涉及自噬的CVD之间的关系,还需要进行很多探索。 摘要:越来越多的证据表明,lncRNA在CVD的背景下对调节自噬起了重要作用。在这篇综述中,我们首先总结了有关lncRNA在心血管自噬和涉及自噬的CVD中所起的作用的当前知识。然后,讨论了设计用于靶向lncRNA来特异性调节患病心脏和血管中自噬的反义寡核苷酸(ASO)的最新发展,主要侧重于不同化学修饰的结构与活性之间的关系以及相关的临床试验。 观点:ASO在心血管药物创新中很有前途。我们希望将来对基于lncRNA的疗法的研究能够克服现有的技术局限性,并为患有自噬性CVD的人们提供帮助。
