摘要

耐多药细菌的出现已成为现代医学中的紧迫问题,这需要开发抗生素的新策略。最近的研究支持这一假设,即抗生素诱导的细菌细胞死亡是由活性氧(ROS)介导的。该假设还强调了抗氧化系统的重要性,抗氧化系统是有助于抗生素耐药性的防御机制。硫氧还蛋白和谷胱甘肽系统是两个主要的硫醇依赖性系统,它们不仅提供抗氧化能力,而且还参与细菌中的各种生物学事件,例如DNA合成和蛋白质折叠。生物学上的重要性使它们成为新型抗生素开发的有希望的靶标。基于该思想,已发现两种细菌硫氧还蛋白还原酶抑制剂依布硒啉和金诺芬可有效抑制缺乏GSH的细菌的生长。依布硒啉和银的最新研究表明,它对同时具有硫氧还蛋白和谷胱甘肽系统的耐多药(MDR)革兰氏阴性细菌具有很强的协同作用。这些具有药物用途的研究由于其众所周知的特性而有望用于快速的临床应用。
