在过去的二十年里,纳米医学领域的发展没有人们普遍期望的那么成功。其主要原因是生物系统极其复杂,包括生物流体的物理化学性质,以及生物化学和生物系统的生理学。纳米粒子的物理化学性质也非常重要。这与新合成的粒子在生物/生物系统中的应用有很大的不同,最近的研究表明。纳米粒子的物理化学性质是由它们的结构和功能设计(核心和涂层材料)预先确定的,并且受到它们与环境(温度、pH、盐、蛋白质、细胞)相互作用的高度影响。由于涂层材料是粒子与环境接触的第一部分,它不仅提供了生物相容性,而且还定义了纳米粒子在生物系统中的行为(如胶体稳定性)和归宿(降解、排泄、积累)。因此,涂层很重要,特别是在生物医学应用中的纳米粒子系统,它必须在复杂的生物流体、细胞和生物环境中完成其任务。本文对不同的纳米涂层材料在生物介质和生活系统中提供胶体稳定性的能力进行了评价。

摘要: Within the last two decades, the field of nanomedicine has not developed as successfully as has widely been hoped for. The main reason for this is the immense complexity of the biological systems, including the physico-chemical properties of the biological fluids as well as the biochemistry and the physiology of living systems. The nanoparticles’ physicochemical properties are also highly important. These differ profoundly from those of freshly synthesized particles when applied in biological/living systems as recent research in this field reveals. The physico-chemical properties of nanoparticles are predefined by their structural and functional design (core and coating material) and are highly affected by their interaction with the environment (temperature, pH, salt, proteins, cells). Since the coating material is the first part of the particle to come in contact with the environment, it does not only provide biocompatibility, but also defines the behavior (e.g. colloidal stability) and the fate (degradation, excretion, accumulation) of nanoparticles in the living systems. Hence, the coating matters, particularly for a nanoparticle system for biomedical applications, which has to fulfill its task in the complex environment of biological fluids, cells and organisms. In this review, we evaluate the performance of different coating materials for nanoparticles concerning their ability to provide colloidal stability in biological media and living systems.
