摘要
内在或获得性化疗耐药是癌症患者成功治疗的主要障碍。多药耐药机制涉及:亲水性药物摄取减少,外排能量依赖性增加,氧化还原状态改变,凋亡途径改变,肿瘤微环境改变等。近年来,开发了几种纳米粒子来克服这些障碍,并且改善药物在病理部位的积聚和释放。在这篇综述中,我们描述了涉及多重耐药性的主要机制和已经提出的针对这一现象的特定方面的纳米载体。

摘要: Intrinsic or acquired chemoresistance represents the main obstacle to the successful treatment of cancer patients. Several mechanisms are involved in multidrug resistance: decreased uptake of hydrophilic drugs, increase of energy dependent efflux, alteration of the redox state, alteration of apoptotic pathways, and modification of the tumor microenvironment. In recent years, several types of nanoparticles have been developed to overcome these obstacles and improve the accumulation and release of drugs at the pathological site. In this review, we describe the main mechanisms involved in multidrug resistance and the nanovehicles which have been proposed to target specific aspects of this phenomenon.
