摘要
目前,世界上许多人都患有慢性丙型肝炎 C. HCV NS5B聚合酶,保守的确定7 HCV基因型被认为是最有前途的目标打击HCV。在过去的十年中,显著的进步已经在新型核苷HCV NS5B聚合酶抑制剂的发现了。一种有效的治愈率高抗HCV药物已经被了批准。此外,相当多的核苷抗丙型肝炎病毒药物在临床试验中正在被评估。本综述的目的是目前核苷HCV NS5B聚合酶抑制剂的发展进展,重点把握药用化合物及其构效关系(SARS)的承诺为未来的药物设计和发现提供指导。

摘要: Nowadays, a large number of people in the world are suffering from chronic Hepatitis C. HCV NS5B polymerase conserved across the identified 7 HCV genotypes is considered to be the most promising target in combating HCV. During the past decade, significant progress has been made in the discovery of novel nucleoside HCV NS5B polymerase inhibitors. A potent anti-HCV drug, sofosbuvir with high cure rates has been approved. Besides, quite a few nucleoside anti-HCV agents are being evaluated in clinical trials. The purpose of this review is to present recent progress in the development of nucleoside HCV NS5B polymerase inhibitors, focusing on lead compounds that hold great promise for medicinal use and their structure-activity relationships (SARs) in order to provide guidance for future drug design and discovery.
