摘要
控制癌症发展和进展的机制被认为是遗传畸形和表观遗传修饰的结果。因此,表观遗传学已经成为一个令人难以置信的和逐步探索的研究领域,以发现更好的肿瘤预防和治疗,特别是三阴性乳腺癌(TNBC)。它占所有浸润性乳腺癌的15-20%,通常具有好斗的组织学特征和不良的临床结果。在三阴性乳腺癌发生的早期,表观遗传异常改变染色质结构,影响细胞的可塑性。它通过获得不受限制的自我更新能力来维持恶性祖细胞的致癌重编程。TNBC的基因组冲动引发突变、拷贝数变化和基因重排,而表观遗传重构包括DNA甲基化、组蛋白修饰和基因表达谱的非编码RNA的修正。目前很明显,表观遗传机制在TNBC的发病、维持和治疗耐药中起着重要作用。虽然TNBC是一种复杂的疾病,难以描述和治疗,但正在进行的遗传学和表观遗传学研究将有助于描述和治疗TNBC。转录组分析的最新进展改变了我们对人类疾病的认识,包括分子医学层面的TNBC。它是吸引人的设想转录组生物标志物,以理解肿瘤行为更容易有关其细胞微环境。了解这些重要的生物标志物和分子变化将增强我们充分治疗TNBC的能力。这篇综述将从分子医学的角度描述表观遗传学的不同方面和转录组学在三阴性乳腺癌发生中的前景,以及它们在诊断、预后和治疗决策中的应用。
