摘要
整合素是跨膜异二聚体蛋白中的一个大家族,是细胞外基质成分的主要受体。整合素最初被认为参与维持细胞粘附和组织的完整性。然而,现在科学家认识到整合素在许多生物过程如细胞存活、增殖、分化、迁移、细胞形态和极性中发挥重要作用。肺细胞能够表达多种整合素异二聚体的排列和组合方式。应用于不同肺病的整合素异二聚体的复杂性和多样性是药物开发面临的主要挑战。本文对在细胞培养中整合素基因敲除小鼠模型的研究进行了全面的概述,并提供了整合素作为呼吸系统疾病(哮喘,肺气肿,肺炎,肺癌,肺纤维化、结节病)的主要治疗靶点的临床试验的最新结果 。

摘要: Integrins are a large family of transmembrane heterodimeric proteins that constitute the main receptors for extracellular matrix components. Integrins were initially thought to be primarily involved in the maintenance of cell adhesion and tissue integrity. However, it is now appreciated that integrins play important roles in many other biological processes such as cell survival, proliferation, differentiation, migration, cell shape and polarity. Lung cells express numerous combinations and permutations of integrin heterodimers. The complexity and diversity of different integrin heterodimers being implicated in different lung diseases present a major challenge for drug development. Here we provide a comprehensive overview of the current knowledge of integrins from studies in cell culture to integrin knockout mouse models and provide an update of results from clinical trials for which integrins are therapeutic targets with a focus on respiratory diseases (asthma, emphysema, pneumonia, lung cancer, pulmonary fibrosis and sarcoidosis).
