神经系统疾病是与神经元和中枢神经系统(CNS)有关的疾病。神经元的任何结构、电、生化和功能异常都可能导致各种类型的疾病,如阿尔茨海默病(AD)、抑郁症、帕金森病(PD)、癫痫、中风等。目前可用的药物是对症的,不能治疗疾病状态。因此,人们迫切需要具有彻底治疗疾病潜力的新型中枢神经系统活性药物。一系列有机小分子正在被探索作为治疗不同神经系统疾病的潜在候选药物。在此背景下,芳基哌嗪已被发现是许多中枢神经系统活性药物中不可或缺的多用途支架和药效团。一些具有芳基哌嗪核的分子已被开发为治疗AD、PD、抑郁症和其他疾病的有力先导。芳基哌嗪核可以在不同的化学结构上选择性取代,为大量衍生物的合成提供了灵活性。在目前的综述文章中,我们已经探索了各种含芳基哌嗪的支架对不同神经系统疾病的作用,包括AD, PD和抑郁症。对有效先导化合物进行了构效关系研究。这篇综述文章可能为设计和合成有效分子作为治疗不同神经疾病的药物提供重要的结构要求。
