摘要
肺癌是当今世界上主要的癌症类型。激酶在介导信号传导途径中起着至关重要的作用,并且它直接控制一些必要的细胞过程。相反,酪氨酸激酶的失调导致致癌转化,不受控制的细胞增殖和肿瘤发生。酪氨酸激酶在肺癌中特别是在非小细胞肺癌(NSCLC)中被大大放松调节。因此,抑制致病性激酶是癌症研究,治疗和护理的突破性进展,可从临床上改善生活质量。在过去的几十年中,各种单一或组合抑制剂已获得美国食品药品监督管理局(FDA)的批准,可在临床上买到,目前,正在进行一些临床前研究并检查激酶抑制剂。但是,在了解激酶抑制剂的机制及其选择性方面仍存在许多空白。在此分析中,我们着眼于一类受体和非受体酪氨酸激酶抑制剂及其在肺癌中的新作用。
