多能干细胞(PSC)是研究发育生物学和神经元疾病的强大工具。 传统的分化方案需要几种中间状态和不同的培养条件,从而无法有效地生成具有未成熟特征的神经元细胞的混合亚型。 通过强制表达神经元转录因子对PSC进行直接编程已显示出高纯度的快速细胞命运测定方法,因为它可以绕开传统培养所需的顺序发育步骤。 在这篇综述中,我们着重于通过各种转录因子将神经元从PSC分化为特定的亚型。 此外,讨论了这种新颖技术的潜在应用和局限性。
