摘要
美洲锥虫病是一种寄生虫感染,通常称为恰加斯病,影响到拉美各国的数百万人。目前,世界卫生组织(WHO)预测,每年发生的死亡人数超过10,000人,国际受感染者人数将增加到700万至800万。通过迁移到非流行世界国家的人们传播病原体克氏锥虫(Trypanosoma cruzi)使其成为一种新发疾病。针对锥虫杀虫药的最有希望的目标可分为三大类:第I组包括主要分子靶标,其被认为是参与寄生虫存活的基本过程的特定酶,主要是Cruzipain,主要抗原寄生虫半胱氨酸蛋白酶。第II组涉及生物途径及其关键特异性酶,例如甾醇生物合成途径,特别是抗氧化防御机制和生物能量学途径。 III组包括存在于寄生虫相关临床形式中的非典型细胞器/结构,其与哺乳动物和与其相关的生物学过程中存在的那些不存在或显着不同。这些可被认为是开发具有额外有效性和比目前使用的治疗剂更少的副作用的药物的潜在靶标。改善宿主和寄生虫目标之间的区别将有助于对抗这种被忽视的疾病。
