肽聚糖是细菌细胞的外骨骼,是保护细胞的重要屏障,其合成的最终步骤是由转肽酶催化的。了解这些在结核分枝杆菌中产生这种大分子的重要酶的结构和功能有助于开发有效的抗结核先导化合物。本文综述了迄今为止在结核分枝杆菌转肽酶方面的实验和计算研究,这些研究已被确认和验证。综述了L,D-和D,D-转肽酶的结构及其功能,并对L、D转肽酶的酶促机制进行了综述。此外,我们根据实验和计算方法提供已知结核病药物对这些酶的生物活性。随着对这些突出靶点的了解不断深入,有助于开发具有新型抑制机制的新药,从而克服当前对结核病新药的需求。
