摘要
背景:无菌的alpha motif (Sam)域是一个小的螺旋蛋白模块,能够发生相同或不同的低聚反应,以及聚合作用,从而形成不同类型的蛋白结构。少数Sam区域参与了病理过程,因此,它们为开发新的潜在治疗途径提供了有价值的靶点。本研究的目的是收集关于Sam区域可以促进疾病发生和进展的不同模式的最新知识。 方法:这篇综述是通过检索文献建立的: a) Sam结构域的结构特性,b) Sam模块介导的相互作用,c)与特定疾病相关的蛋白质中Sam结构域的存在。 结果:Sam结构域在癌症、肾脏疾病、白内障等疾病中起重要作用。通常病理与直接定位于Sam区域的突变有关,这些突变改变了它们的稳定性和/或影响了对适当的蛋白质功能至关重要的相互作用。在少数疾病中,Sam motif起到一种“侧作用”,通过增强不同蛋白域的作用来协同病理事件。 结论:考虑到Sam领域在多种疾病中的作用,需要更多的努力和新的药物发现运动来发现针对Sam领域的小分子和/或多肽。这类化合物可能是构建治疗不同疾病的新策略的基础。
