与从头药物发现方法相比,药物重新利用是加快新型药物发现和开发过程的安全且成功的途径。药物替代用途使用FDA批准的药物和在临床试验中失败的药物,这些药物具有有关潜在毒性,制剂和药理学的详细信息。信息学,基因组学和生物科学方面的技术进步是药物重用在确定现有药物的次要适应症方面的重大成功的原因。改变用途的药物在填补潜在抗生素发现方面起着至关重要的作用。由于对现有药物的多药耐药性,细菌感染已成为日益增长的全球公共卫生威胁。这就迫切需要开发能够有效抵抗多药耐药细菌感染(MDRBI)的新型抗生素。本综述描述了在2016-2017年间,药物改用在抗生素开发中的关键作用,以及最近FDA批准的抗生素,管道抗生素以及各种FDA批准的抗癌,抗真菌药的抗菌特性的详细信息。抗高脂血症,抗炎,抗疟疾,抗寄生虫,抗病毒,遗传疾病,免疫调节剂等。此外,鉴于与现有抗生素的联合疗法,将探讨其对MDRBIs的潜在影响。当前的综述可能为开发满足当前需求的快速,安全,有效和新颖的抗生素提供必要的数据,并建议通过重新使用现有药物来抑制MDRBI的有效含义方面的敏锐度。
