大肠癌(CRC)是全球范围内与癌症相关的死亡的主要原因。 CRC风险增加与肠道菌群改变,短链脂肪酸(SCFA)产生减少有关。在人类结肠中产生的SCFA是未经消化的膳食纤维和淀粉细菌发酵的主要产物。结肠细胞使用乙酸,丙酸酯和丁酸酯这三种主要的SCFA作为能源,而转化的CRC细胞主要经历有氧糖酵解。与正常结肠细胞相比,CRC细胞对SCFA的敏感性增强,因此表明它们在细胞体内平衡中起着重要作用。因此,通过改变微生物群来控制肠道中的SCFA水平已成为一种潜在的CRC预防/治疗策略。近年来,人们对了解SCFA在CRC细胞中的作用机制的兴趣有所增加。几种SCFA转运蛋白如SMCT-1,MCT-1和水通道蛋白已被确定为肠道细胞中的主要跨膜转运蛋白。最近,显示乙酸盐促进MCT-1的质膜重新定位并触发葡萄糖代谢的变化。 SCFAs诱导CRC细胞凋亡,并发现了进一步的机制,包括溶酶体膜通透化与线粒体功能障碍和降解有关。在这篇综述中,我们将讨论CRC细胞转运SCFA的最新知识及其对CRC代谢和存活的影响。也将解决通过操纵结肠微生物群增加SCFA产生对CRC预防/治疗的影响。
