摘要
线粒体是人类细胞唯一的半自主细胞器,不仅在能量生产中而且在细胞凋亡,代谢和细胞信号传导中都起着非常重要的作用。它们也是ATP生产过程中ROS和RNS自由基的主要生产者。过量的这些自由基可能损害线粒体DNA以及蛋白质组,导致错误折叠蛋白质的积累,这可能证明它们的功能是有害的并且已知参与疾病病理学。为了维持健康的蛋白质组,线粒体已经作为半自主方式的质量控制机制发展,细胞蛋白质如蛋白酶和热休克蛋白被用于质量控制。本综述讨论了在外膜或内膜以及膜间隙操作的线粒体蛋白质质量控制的各个方面。已经讨论了涉及线粒体解折叠蛋白应答的各种途径以及它们在癌症和各种神经退行性疾病中的意义

摘要: Mitochondria are the sole semiautonomous organelles of the human cell and play a very important role in not only energy production but also in apoptosis, metabolism and cell signaling. They are also known to be major producers of ROS and RNS free radicals during ATP production. These free radicals in excessive amount may damage the mitochondrial DNA as well as proteome resulting in accumulation of misfolded proteins which may prove deleterious to their functioning and are known to be involved in disease pathology. To maintain healthy proteome, mitochondria have developed as quality control machinery in semiautonomous manner, where cellular proteins such as proteases and heat shock proteins are used for quality control. The present review discusses various aspects of mitochondrial protein quality control operating at outer or inner membrane as well as intermembranal space. The various pathways involved in mitochondrial unfolded protein response have been discussed along with their implications in cancer and various neurodegenerative diseases.
