摘要
Autotaxin(ATX)已成为一个在LPA相关疾病中有巨大的药理学和药物化学兴趣的充满吸引力的目标,至今许多有机小分子已经被探索来用作潜在的ATX抑制剂。作为各种鉴别新型有效的ATX抑制剂的有效协助,生物信息学同样也起着十分重要的作用。特别是,由于它便捷的大数据库,先进的技术和令人鼓舞的事实,虚拟筛选(VS)最近受到了越来越多的关注,从VS中发现的几种化合物,已经达到了市场或进入临床试验的发展阶段。不同的技术和工作流程已经以优先可能被点击的目标在文献中报告。在这篇综述中几个部署的虚拟筛选策略对新型强效ATX抑制剂的鉴定进行了描述。

摘要: Autotaxin (ATX) has become an attractive target with a huge pharmacological and pharmacochemical interest in LPA-related diseases and to date many small organic molecules have been explored as potential ATX inhibitors. As a useful aid in the various efforts of identifying novel effective ATX inhibitors, in silico methods can serve as an important and valuable tool. Especially, Virtual Screening (VS) has recently received increased attention due to the large datasets made available, the development of advanced VS techniques and the encouraging fact that VS has contributed to the discovery of several compounds that have either reached the market or entered clinical trials. Different techniques and workflows have been reported in literature with the goal to prioritize possible potent hits. In this review article several deployed virtual screening strategies for the identification of novel potent ATX inhibitors are described.
