摘要
缺口信号途径是一种高度的保护系统,它控制了胚胎细胞命运的决定和通过影响相邻细胞的交流来控制成熟干细胞的维持。这种途径与由于增加的细胞增殖和肿瘤血液灌注除此之外还有抑制细胞凋亡引起的各种癌症的发展有关联。医药代表压制过于活跃的缺口信号也许有利于癌症病人的治疗。这些定向的疗法比较常规的抗癌疗法包括减少对正常细胞的影响有更多的有利条件。在这个综述中,我们探索在癌症中定向的缺口信号途径的基本原理和调查研究的策略旨在阻止该通路。

摘要: The Notch signaling pathway is a highly conserved system that controls embryonic cell fate decisions and the maintenance of adult stem cells through affecting communication between adjacent cells. The pathway is linked to the development of various cancers owing to increased cell proliferation and tumor blood perfusion in addition to inhibition of apoptosis. Pharmaceutical agents that suppress overactive Notch signaling may be of benefit in the treatment of patients with various cancers. These targeted therapies confer several advantages over conventional anticancer therapies including reduced deleterious effects on normal cells. In this review, we explore the rationale for targeting the Notch signaling pathway in cancer along with different investigational strategies designed to block the pathway.
