游离脂肪酸(FFA)除了作为一个重要的燃料资源和生物膜结构成分,也显示了多样的作用,包括受体信号的调整和基因表达的调整。FFA能激活胰腺 β 细胞,免疫细胞,白色脂肪组织,肠和其他一些组织中特定的G蛋白偶联受体(GPCR),因此在维持代谢平衡上起重要作用。游离脂肪酸受体2(FFAR2)也被称为属于GPCR类的GPR43,它参与了许多重要的生命活动。FFAR2是被短链脂肪酸(SCFA)激活的,如乙酸,丙酸,丁酸。SCFA是由未在上部消化道被消化而进入结肠的大纤维物质被细菌发酵形成,它在免疫调整和代谢平衡中起关键作用。FFAR2和其他游离脂肪酸受体被视为人体的营养传感机理的关键部分,对这些受体的针对手段被认为对糖尿病和其他 代谢紊乱有特效。这篇综述诣在总结我们对FFAR2生物学的理解的现状并强调了它在代谢平衡中的作用。

摘要: Besides being an important source of fuel and structural components of biological membranes, free fatty acids (FFAs) are known to display a wide variety of roles that include modulation of receptor signaling and regulation of gene expression among many. FFAs play a significant role in maintaining metabolic homeostasis by activating specific G-Protein Coupled Receptors (GPCRs) in pancreatic β cells, immune cells, white adipose tissue, intestine and several other tissues. Free Fatty acid receptor 2 (FFAR2) also known as GPR43 belongs to this group of GPCRs and has been shown to participate in a number of important biological activities. FFAR2 is activated by short-chain fatty acids (SCFAs) such as acetate, propionate and butyrate. SCFAs are formed in the distal gut by bacterial fermentation of macro-fibrous material that escapes digestion in the upper gastrointestinal tract and enters the colon and have been shown to play vital role in the immune regulation and metabolic homeostasis. FFAR2 and other free fatty acid receptors are considered key components of the body’s nutrient sensing mechanism and targeting these receptors is assumed to offer novel therapies for the management of diabetes and other metabolic disorders. This review aims to summarize the current state of our understanding of FFAR2 biology with a particular focus on its role in metabolic homeostasis.
