摘要
外泌体是介导细胞间通讯的纳米载体,对正常生理功能至关重要。然而,呈指数级增长的报告已经将其失调的释放与包括癌症在内的各种病理学联系起来。在癌症中,从间质重塑到转移,肿瘤细胞绕过免疫监控并显示出药物抗性,已证实其通过肿瘤衍生的外泌体介导。由于它们在癌症致病性中的作用,基于外泌体的策略在治疗方案中具有巨大的潜力。这些策略包括将外来体用作药物载体或免疫治疗剂,这需要先进的纳米技术来分离和表征外来体。相反,对外来体机制的药理学抑制超过了纳米技术的要求,因此成为癌症治疗的一个重要前景。在这方面,研究人员目前正试图剖析分子途径,以揭示促进肿瘤源性外泌体释放的关键调节蛋白的参与。随后,正在进行针对这些蛋白质的各种分子的筛选,最终降低外源体诱导的癌症致病性。然而,它们的临床翻译需要更广泛的研究。在这里,我们全面回顾了调节癌症中外泌体释放的分子机制。此外,我们还深入了解了突出各种药物作为外泌体阻断剂的作用的关键发现,这将为癌症治疗中的药物开发提供新的途径。
