摘要
雷帕霉素(rapamycin, mTOR)的作用靶点是细胞代谢和生长的关键调控因子。mTOR以两种不同的多蛋白复合物mTORC1和mTORC2的形式整合细胞能量、营养和激素信号,调节细胞代谢稳态。在2型糖尿病(T2DM)中,病理条件和末端器官并发症可归因于异常的mTOR。大量证据表明,mTORC1-p70S6激酶1 (S6K1)和mtorc2蛋白激酶B (AKT)这两种mTORC1-p70S6激酶信号通路在胰岛素敏感性中发挥关键作用,它们的功能障碍有助于T2DM的发生。本文综述了我们目前对mTOR信号通路在T2DM及其相关并发症中的作用的认识,以及mTOR抑制剂在T2DM治疗中的潜在应用。
