摘要
本文综述了脂基纳米载体(LNCs)在药物活性物质非肠道输送中的应用。首先,从制备、表征和稳定性等方面介绍了两代纳米脂质纳米颗粒(SLNs)和纳米脂质载体(NLCs)。虽然通过非肠道给药使用LNCs已显示出许多益处,但其使用受到调理作用的限制,调理作用是一种导致其半衰期短(3-5分钟)的免疫过程。因此,许多策略被讨论,以实现“隐形”系统适合肠外给药。综述了脂质纳米颗粒在天然化合物、合成药物和遗传物质输送方面的需求和应用。最近,由于mRNA疫苗在抗击Covid-19大流行方面的众多益处,后者的应用受到了极大的关注。

摘要: This review describes the use of lipid-based nanocarriers (LNCs) for the parenteral delivery of pharmaceutical actives. Firstly, the two generations of LNCs, namely solid lipid nanoparticles (SLNs) and nanostructured lipid carriers (NLCs), are explained in terms of preparation, characterization and stability. Although the use of LNCs through parenteral administration has shown many benefits, their use is limited by opsonization, an immune process that causes their short half-life (3-5 min). Therefore, many strategies are discussed to realize “stealth” systems suitable for parenteral administration. The requirements and applications of parenteral lipid nanoparticles are reviewed for the delivery of natural compounds, synthetic drugs and genetic materials. Recently, the latter application has been of remarkable interest due to the numerous benefits of mRNA vaccines to fight the Covid-19 pandemic.
