岩藻黄质对脑室内链脲佐菌素 (ICV-STZ) 诱导的实验大鼠认知障碍的神经保护作用

背景:阿尔茨海默病(AD)是一种神经系统疾病,其特征是由氧化应激、神经炎症、神经递质水平变化和 Aβ(1-42)斑块过度沉积引起的记忆和认知功能丧失。岩藻黄素是一种类胡萝卜素,具有潜在的抗氧化、抗炎和神经保护作用。目的:在本研究中,岩藻黄质被用作脑室链脲佐菌素(ICV-STZ)诱导的认知障碍实验模型的保护策略。 方法:STZ 在第 1 天和第 3 天交替注射两次 ICV (3 mg/kg),并使用 Morris 水迷宫和高架十字迷宫评估 Wistar 大鼠的记忆分析。低 50 mg/kg 的岩藻黄质,口服和高剂量 100 mg/kg,口服给药14天。第29天处死所有动物,分离后的脑海马组织用于生化(MDA、亚硝酸盐、GSH、SOD和过氧化氢酶)、神经炎症(TNF-α、IL-1β和IL-6)、神经递质(ACh、 GABA 谷氨酸盐)、Aβ(1-42) 和 Tau 蛋白测量。 结果:注射 STZ 的大鼠表现出明显的学习和记忆障碍,氧化应激(MDA、亚硝酸盐)增加,抗氧化防御(GSH、SOD 和过氧化氢酶)降低,促进细胞因子释放和神经递质水平变化。然而,岩藻黄质改善了认知功能,恢复了抗氧化水平,剂量依赖性地减少了炎症标志物,并恢复了神经递质浓度。 结论:目前的研究结果表明,岩藻黄质可能是通过抗氧化、抗炎和神经保护机制以及抑制乙酰胆碱酯酶 (AChE) 酶活性、Aβ(1-42) 积累和τ蛋白。
