与衰老和神经退行性疾病相关的脑功能缺陷从现有治疗选择中受益不大,这表明缺乏对病理机制的了解。阿尔茨海默氏病(AD)是一种几乎无法治疗的疾病,与年龄神经系统退化有关,只有对症治疗可用,而且能导致疾病进展的药剂还很遥远。AD中磷酸二酯酶(PDE)表达的改变和环核苷酸信号通路的解除引发了靶向AD中环核苷酸信号通路的新思路。以环核苷酸为靶点作为胞内信使似乎是一种可行的方法,在大脑中某些生物过程和控制实质。然而,环核苷酸的合成、执行和/或降解与认知缺陷密切相关。在认知方面,环核苷酸(cAMP和cGMP)在记忆的不同阶段,包括基因转录、神经发生、神经元回路、突触可塑性和神经元存活等,都有着不可避免的作用。这些基本认知过程的损伤是AD的表现,这表明cAMP/cGMP信号在AD人群中起着至关重要的作用。磷酸二酯酶抑制剂是唯一的一套酶,促进水解和降解cAMP和cGMP,从而保持其最佳水平,启动作为一个有趣的目标探索。本文综述了PDE抑制对AD的生理状态、病理进展和神经生物学标志物的神经保护作用和实质性影响,与cAMP和cGMP的强度一致。
