摘要
背景:泛素-蛋白酶体通路对所有细胞过程都至关重要,因此是研究和开发癌症治疗新策略的关键靶点。除此之外,由于翻译错误或翻译后修饰缺陷,约30%的新合成蛋白无法达到其最终构象;因此,它们也被泛素-蛋白酶体途径快速清除。 目的:本文简要介绍了在细胞培养实验和临床试验中涉及到调控泛素蛋白酶体通路的新分子机制以及对蛋白酶体抑制剂的耐药机制的最新发现。 结果:由于癌细胞有较高的增殖率和更容易翻译错误,他们需要泛素-蛋白酶体途径的选择优势和持续增殖。因此,靶向泛素-蛋白酶体途径的药物对于治疗血液病和实体癌都是很有前途的药物。 结论:许多蛋白酶体抑制剂被批准并用于治疗晚期和复发的多发性骨髓瘤。不幸的是,耐药机制可能在蛋白酶体抑制剂治疗开始的几天内迅速发生发展,这可能是由于在选择性药物压力下的固有或获得性耐药机制所致。然而,全面了解导致蛋白酶体抑制物耐药的机制,最终将有助于设计和开发新的策略,包括治疗一些癌症的新药和/或药物组合。
