背景与目的:丝虫病是一种被忽视的热带病,是由被蚊虫叮咬传播的丝虫感染引起的。在全球范围内,有1.2亿人被感染,近4000万人因腿部严重肿胀((病)或阴囊(鞘膜积液)等并发症而致残。目前的治疗方法(伊维菌素,二乙基卡巴马嗪)对成人寄生虫的作用有限,并产生副作用。因此,迫切需要寻找新的抗孝顺剂。在最近的文献中,已经报道了许多有关纯分子的抗丝活性的研究。本研究描述了针对淋巴丝虫病的有效抗丝虫化合物的现状。 方法:从开始到2019年9月,通过参考教科书和科学数据库(SciFinder,PubMed,Science Direct,Wiley,ACS,SciELO,Google Scholar和Springer等)对天然存在的和合成的抗丝虫化合物进行文献检索。 结果:据报道,许多化合物在导致淋巴丝虫病的成虫和微丝虫病形式的寄生虫中均具有抗丝虫活性。在计算机上对活性抗丝虫化合物(配体)进行的研究表明,淋巴丝虫病的蛋白质靶点(海藻糖6磷酸磷酸酶,胸苷酸合酶等)具有分子相互作用,并支持体外结果。 结论:关于体外抗丝虫研究,有证据表明天然和合成产品可作为抗丝虫剂开发的基本支架。可以进一步进行最有效的抗丝虫化合物的优化,然后进行体内研究。
