摘要
背景: 目前癌症治疗的一些挑战和并发症是化疗引起的周围神经病变(CIPN)和与这种情况相关的神经痛。许多主要的化疗药物可引起神经毒性,对免疫系统有显著的调节作用,并常伴有各种不良反应。最近的证据表明,在化疗期间,神经系统和免疫系统之间会发生串音;因此,一个新出现的概念是,神经炎症是CIPN的主要机制之一,这已被趋化因子的上调所证实。趋化因子最初被认为是外周免疫细胞转运的调节因子,在中枢神经系统的神经元和胶质细胞上也有表达。 目的: 在这篇综述中,我们收集的证据表明趋化因子是CIPN疼痛信号的潜在介质和贡献者。趋化因子及其受体CX3CL1/CX3CR1、CCL2/CCR2、CXCL1/CXCR2、CXCL12/CXCR4、CCL3/CCR5在CIPN病理状态下表达改变,趋化因子受体拮抗剂可减轻神经性疼痛行为。 结论:通过对趋化因子介导的通讯机制的了解,我们可能发现趋化因子靶点可以作为治疗CIPN的新策略。
